The project starts after confirmed by the client, and the final delivery antibody will be checked through phosphorylated peptides and non phosphorylated peptide double testing. Phosphorylation antibody can specifically identify the phosphorylated amino acid sites. Phosphorylation antibody can be used to distinguish between the existence of protein phosphorylation and non phosphorylated form, can be used for qualitative and quantitative analysis of protein phosphorylation. In the project, animal immune serum collected is purified through phosphorylated peptides affinity chromatography column firstly, then the preliminary purification antibody is absorbed with non phosphorylated peptide affinity chromatography column, so the final delivery phosphorylation antibody is confirmed with the BSA crosslinking of phosphorylated peptide and non phosphorylated peptide double test to make sure the phosphorylation antibody delivery quality.
Phosphorylation polyclonal antibody preparation (rabbit)
Firstly, Design and synthesis of phosphorylated peptide, synthesis 10 mg of phosphorylated peptides and 5 mg of phosphorylated peptides.
Secondly, Peptide coupling, synthesis peptides coupling with carrier protein (KLH, BSA), 2 ~ 3 mg peptide (non phosphorylated peptide and phosphorylated peptides) will be provided.
Thirdly, Immune rabbit, serum before the immune, immune four times and serum. 1~ 2 ml serumbefore the immune will be provided.
Fourthly, Small-scale purified, serum sample collection and detection. 2 ~ 5 mg phosphorylation antibody and testing result by ELISA will be provided.
Fifthly, Antigen affinity purification, preparation phosphorylation affinity purification column, non phosphorylated peptides affinity column to remove cross reaction. Project report will be provided.
Sixthly, Detect titer by ELISA.
Phosphorylation of monoclonal antibody preparation (mouse)
Firstly, Design and synthesis of phosphorylated peptide, synthesis 10 mg of phosphorylated peptides and 5 mg of phosphorylated peptides.
Secondly, Peptide coupling, synthesis peptides coupling with carrier protein (KLH, BSA), 2 ~ 3 mg peptide (non phosphorylated peptide and phosphorylated peptides) will be provided.
Thirdly, Animal immunization and detection, immune mice and tested by ELISA with phosphorylation and non phosphorylated peptide. 5 mg modified peptides, 3 mg undecorated peptides, antibodies and 50 ul serum before immune will be provided.
Fourthly, Cell fusion, phosphorylated peptide showed the immune response, mice spleen cells and myeloma cells for cell fusion. 2-5 strains hybridoma, 3-8 mg purified antibody (2 strains) and ELISA test results will be provided.
Fifthly, Proliferation and selection, detect the positive cloning by ELISA and the subcloning. Subtype identification results and complete project report will be provided.
Sixthly, Ascites preparation and purification, purification of Protein G.
Seventhly, Detect titer by ELISA.
